چکیده

NADH:ubiquinone reductase (complex I) of the mitochondrial inner membrane respiratory chain binds a number of mitochondrial matrix NAD-linked dehydrogenases. These include pyruvate dehydrogenase complex, a-ketoglutarate dehydrogenase complex, mitochondrial malate dehydrogenase, and &hydroxyacylCoA dehydrogenase. No binding was detected between complex I and cytosolic malate dehydrogenase, glutamate dehydrogenase, NAD-isocitrate dehydrogenase, lipoamide dehydrogenase, citrate synthase, or fumarase. The dehydrogenases that bound to complex I did not bind to a preparation of complex I1 and 111, nor did they bind to liposomes. The binding of pyruvate dehydrogenase complex, a-ketoglutarate dehydrogenase complex, and mitochondrial malate dehydrogenase to complex I is a saturable process. Based upon the amount of binding observed in these in vitro studies, there is enough inner membrane present in the mitochondria to bind the dehydrogenases in the matrix space. The possible metabolic significance of these interactions is discussed.

The effect of 6-deoxyclitoriacetal from Clitoria macrophylla Wall. on rat liver mitochondrial respiration and ATPase activity

A Thai plant Clitoria macrophylla Wall. was claimed to possess some pharmacological activities. The phytochemical studies reported a rotenoid compound, 6-deoxyclitoriacetal, which exhibits the cytotoxic effect in several cell line experiments. This report was to study the effect of 6-deoxyclitoriacetal on rat liver mitochondria, which may associated to its cytotoxic phenomena. Mitochondrial sus...

Investigation of NADH Binding, Hydride Transfer, and NAD+ Dissociation during NADH Oxidation by Mitochondrial Complex I Using Modified Nicotinamide Nucleotides

NADH:ubiquinone oxidoreductase (complex I) is a complicated respiratory enzyme that conserves the energy from NADH oxidation, coupled to ubiquinone reduction, as a proton motive force across the mitochondrial inner membrane. During catalysis, NADH oxidation by a flavin mononucleotide is followed by electron transfer to a chain of iron-sulfur clusters. Alternatively, the flavin may be reoxidized...

Multiple Energy Conservation Bypasses in Oxidative Phosphorylation of Plant Mitochondria

The electron transport chain (ETC) in the inner mitochondrial membrane of most eukaryotes consists of four large protein complexes, the rotenone-sensitive NADH dehydrogenase (complex I), succinate dehydrogenase (complex II), cytochrome bc1 complex (complex III) and cytochrome c oxidase (complex IV). The electron transport activity of complexes I, III, and IV (the latter two often jointly called...

Resveratrol Directly Binds to Mitochondrial Complex I and Increases Oxidative Stress in Brain Mitochondria of Aged Mice

Resveratrol is often described as a promising therapeutic molecule for numerous diseases, especially in metabolic and neurodegenerative disorders. While the mechanism of action is still debated, an increasing literature reports that resveratrol regulates the mitochondrial respiratory chain function. In a recent study we have identified mitochondrial complex I as a direct target of this molecule...
